Clinical Significance:
p16 (CDKN2a/INK4a) is an important tumor-suppressor gene, involved in the p16/cyclin-dependent kinase/retinoblastoma gene pathway of cell cycle control. In high-risk HPV infection (particularly in the uterine cervix and head and neck cancer), the E7 HPV protein can inactivate pRB interfere with the pRB regulatory circuit, leading to overexpression of p16. Overexpression of p16 can be used as a surrogate marker for high-risk HPV infection, although HPV infection is not the sole cause of p16 overexpression. It appears to have prognostic significance of head and neck tumors.
